幅広い温度帯で高い活性を維持　

アジレント(旧ストラタジーン)の新製品 AffinityScript Multi Temperature Reverse TranscriptaseはMMLVをベースにしていますが、複数の変異を導入することで、従来の逆転写酵素に比べて幅広い反応温度帯で高い活性が得られるようになりました。図1にあるように、本製品では42～55℃の間で80%以上の高い活性を維持することができます。これに対して競合他社製品では、高い活性を維持できる温度帯が狭く、特に55℃以上や40℃以下では酵素活性が著しく低下します。また、本製品では酵素活性の半減期が長いため、高温での逆転写反応でも十分な量のcDNA合成が可能になっています。本製品では55℃での半減期が35分であるのに対して、競合他社製品では20分以下になっています。

競合他社製品に比べて、AffinityScript Multi Temperature Reverse Transcriptaseでは、より幅広い温度帯で高い活性を維持できています。

このように本製品では、幅広い温度帯で高い収量が得られるため、ランダム・ヘキサマーによる室温での逆転写反応だけでなく、特異的なプライマーを用いた逆転写反応や、GCリッチや二次構造等の難しい配列からの逆転写で、高温での反応が必要な場合にも有効です。図2　少量のRNAからでも高感度で完全長cDNAを合成さらに、使用するRNAの長さに関係なく、十分な量のcDNAの合成が幅広い温度帯で可能なため、反応に合わせて逆転写酵素を変える必要がなく、本製品だけでほぼ全ての逆転写反応を賄うことが出来ます。

少量のRNAからでも高感度かつ十分な収量での完全長cDNA合成

AffinityScript Multi Temperature Reverse Transcriptaseでは、テンプレートとプライマーへの酵素の結合能力（Affinity）が格段に向上しています（MMLVの10倍）。このため、少量のRNAからでも高感度かつ高収量での完全長cDNA合成が可能です。

 図2では、100ng、10ng、1ngのヒトの骨格筋 RNAから50℃で逆転写反応をした後、アジレント(旧ストラタジーン)の高感度PCR酵素 PicoMaxx High Fidelity PCR System (#600420)を用いてPCRを行い、ネブリン遺伝子の5’末端部の0.6kbの領域を増幅しました。他社製品では検出されなかった1ngのRNAからのRT-PCR産物（赤丸部）もアジレント(旧ストラタジーン)製のAffinityScript Multi Temperature Reverse Transcriptaseでは増幅が確認されています。このことから、本製品では、少量のRNAからでも20kbもの長い完全長cDNAの合成が可能であることが示されました。
